Rapid Analysis of Psychotropic Drugs and Metabolites in Urine Using Thermal Desorption Electrospray Ionization Mass Spectrometry

In this study, we developed a rapid analytical method for detecting psychotropic drugs and their metabolites in urine samples using Thermal Desorption Electrospray Ionization Mass Spectrometry (TD-ESI/MS). The misuse of psychotropic drugs represents a significant public health concern, requiring simple yet accurate analytical methods for effective monitoring. We applied the TD-ESI/MS technique, which offers advantages of high sensitivity and selectivity with minimal sample preparation, to analyze urine samples for psychotropic substances. The method was optimized for various therapeutic monitoring drugs and their metabolites that are commonly prescribed in treatment programs. Thermal desorption conditions were adjusted to enhance signal intensity while preserving compound stability. Multiple reaction monitoring (MRM) parameters were optimized for specificity, and method validation confirmed good linearity, accuracy, and recovery. When applied to forensic urine samples, the method successfully distinguished positive cases from blank controls, with MRM chromatograms confirming reliable identification. The system enables rapid analysis in under 1 minutes per sample with minimal preparation, making it well-suited for therapeutic drug monitoring. This approach holds promise for applications in clinical toxicology, forensic analysis, and compliance monitoring.
