Comparison of Amino Acid Analyzer and Triple Quadrupole LC-MS/MS for the Analysis of Biological Free Amino Acids

In the Technical Department at Tottori University, I am responsible for the maintenance and management of shared mass spectrometry equipment as well as performing analytical requests. While strategic facility development and operation plan dictate the renewal and upkeep of university-wide shared facilities, budget constraints have hindered the timely update of aging equipment, making continued use challenging. Since these facilities are utilized by numerous researchers and students, it is essential to implement measures to ensure continued service, whether by using other institutions' equipment or substituting alternative analytical instruments.
Our amino acid analyzer, introduced before 2007, has been serving as a shared facility but aging has made accurate analysis impossible for several amino acids. Consequently, we explored the feasibility of replacing traditional amino acid analysis performed by this equipment with the university's triple quadrupole LC-MS/MS system (LC-MS/MS). This report details the optimization of LC-MS/MS analysis conditions using 26 amino acid standards, the determination of quantifiable concentration ranges for each amino acid, and the analysis of tryptophan in bread wheat seeds as a real sample.
