Medium-molecular-weight Metabolomics Platform with a Focus on Peptides, Developed Based on Capillary Electrophoresis-Mass Spectrometry

Metabolomics targets low-molecular-weight compounds (LCs) typically in the range of 50–1,000 m/z, whereas proteomics usually scans above 10,000 m/z. This implies the existence of a yet-unexplored realm of medium-molecular-weight compounds (MCs) that have not been fully targeted by either omics technologies. We developed a novel high-sensitivity platform based on capillary electrophoresis coupled with a high-resolution mass spectrometry (CE-HRMS) for a comprehensive analysis of MCs with a focus on peptides.
Our large-volume dual preconcentration by isotachophoresis and stacking (LDIS) approach increased the injection volume by 60-fold more than the standard volume, thereby enhancing detection sensitivity by approximately 50-fold more than the original method in CE-HRMS. We applied this platform to analyze the plasma samples from hyperlipidemia patients and healthy controls. As a result, a total of 1,125 peaks were detected including 42 functional peptides annotated using our reference library, 984 peptides identified by PEAKS 12 and 99 unknown peaks. Our medium-molecular-weight metabolomics platform paved the way for the discovery of many novel compounds and yet-unknown metabolic interactions.
