Efficient Exosome Enfichment from Urine Usinf Polyethylene Glycol

This study optimizes a polyethylene glycol (PEG)-based method for isolating high-purity urinary exosomes, overcoming limitations of traditional techniques. Various PEG concentrations, incubation times, and centrifugation conditions were tested to maximize yield and purity. Purification efficiency was evaluated using size-exclusion chromatography (SEC) and high-performance liquid chromatography (HPLC), confirming exosome enrichment. Characterization involved BCA protein assays, Western blot, and scanning electron microscopy (SEM). High-resolution orbitrap mass spectrometry (HRMS) analyzed metabolic differences between urine and exosomes. The optimized method offers a scalable, cost-effective approach for exosome isolation, advancing non-invasive diagnostics and biomarker discovery. Future work will refine purification techniques and validate clinical biomarkers.
