A Novel 18O-Labeled Water Approach for Investigating Intracellular Nucleotide Synthesis Kinetics

Nucleotide metabolism remains challenging to quantify directly. We developed a novel 18O-labeling approach to measure ATP and NTP turnover using H218O incorporation. Cells cultured in 18O-enriched medium rapidly incorporated 18O into ATP via phosphate cycling, with ATP reaching 95% labeling in 6 hours, while other NTPs exhibited distinct kinetics. This method enables cancer-specific metabolic profiling and assessment of drug effects. Our findings highlight localized ATP turnover cycles, providing insights into cellular energy metabolism and therapeutic targeting. Future work will explore regulatory mechanisms and applications in complex biological systems.
