Metabolic signatures of prenatal exposure to 'Cocktails' of benzotriazoles and benzothiazoles and its health implications

In this prospective study of 158 pregnant women in Wuhan, China, we investigated the metabolic alterations associated with prenatal exposure to benzotriazoles and benzothiazoles (BTs). Urinary BT concentrations were repeatedly measured across three trimesters to provide a robust estimate of exposure. High-throughput targeted metabolomics with broad coverage and high accuracy was conducted on third-trimester urine samples to characterize the metabolic profile. We first identified metabolites associated with combined (cocktail) BTs exposure and then examined pairwise associations with individual BTs. A total of 44 metabolites were identified as perturbed biomarkers of cocktail BTs exposure (VIP > 1.2). Subsequent analysis revealed positive associations between BT exposure and oxidative stress-related metabolites and negative associations with metabolites involved in neuronal function. The shared metabolic signatures among BTs, observed in the co-occurrence network analysis, may partly reflect the correlations within the mixture exposures. These findings provide insight into potential mechanisms linking prenatal BT exposure to pregnancy complications and neurodevelopmental risks. Additionally, the analytic framework—integrating mixture and single-pollutant models with omics data—offers a useful approach for future exposome epidemiology studies focused on complex chemical mixtures.
