Development of a Non-Target Analysis Method for Isomeric Peptides in LC-IM-TOFMS

In general, proteins in vivo are synthesized from L-amino acids and composed of L-amino acid residues. However, the existence of isomerized proteins containing isomerized D-amino acid and iso-form residues in vivo has been reported. The analysis of isomerized proteins is in biological systems is an essential and important task. Here, we developed LC-ion mobility-mass spectrometry (LC-IM-MS) as a non-target analysis method for isomeric peptides. The measurement were performed on the trypsin-digested egg white lysozyme. The analytical results suggested that isolation of aspartic acid residues may have occurred in the T12 peptide after accelerated treatment as previously reported. In addition, isomerization of T5 peptides by accelerated treatment was also suggested. This isomerization has not been reported previously, indicating the effectiveness of this method as a non-target analysis. In addition, we developed the method in order to numerically analyze isomerization states. Using the developed method, the isomerization of peptides and the isomerization point were successfully identified.
