Rapid Quantification of Free Aromatic Amines and Related Metabolites in Human Urine Using LC-MS/MS

Certain aromatic amines (AAs) have been classified as human carcinogens by the International Agency for Research on Cancer (IARC). IARC classifies several AAs as Group 1 human carcinogens, with exposure primarily linked to bladder cancer.
Here, we present a rapid and high-sensitivity liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of AAs, related metabolites and internal standard in urine, achieving analysis in under five minutes.
The method demonstrates excellent linearity (r² > [0.995]), with a linear dynamic range of 0.01- 10 ppb. Compared to existing GC-MS approaches, this method eliminates derivatization, reduces analysis time, and improves throughput. The approach enables efficient biomonitoring of AAs for exposure assessment and cancer risk evaluation.
