Effect of Iprodione on Adult Female and Male Zebrafish based on Hormone and Lipidome Analysis using LC-MS/MS

Iprodione is dicarboximide fungicide which was banned in the European Union as of March 5, because of its hepatotoxicity and carcinogenicity. Many studies revealed that iprodione demonstrates acute toxicity to fish such as impairing reproduction and development. However, detailed studies aimed to reveal the mechanism of iprodione toxicity using instrumental analysis are still developing. Despite the possibility of male hormone disturbance potential of iprodione, few studies focused on quantitative analysis of hormones.
This study aimed to discover the detailed toxic mechanism of iprodione to zebrafish using quantitative analysis of hormones with LC-MS/MS. To confirm sexual hormone dysfunction, male and female fishes were divided and analyzed respectively. Using LC-MS/MS multiple reaction monitoring (MRM) mode, and a total of 34 hormones were optimized using derivatization using dansylation. Since hormone synthesis is closely related to lipidome such as cholesterol, lipidome analysis was also conducted using LC-MS/MS. A total of 303 lipids were optimized including 135 phospholipids, 61 sphingolipids, 66 neutral lipids, and 41 small lipids such as carnitine and free fatty acid. This research would provide crucial information to determine the toxicity of iprodione, and its regulation.
