Establishment of technology for single-cell tissue metabolome imaging analysis

Cells exhibit diverse metabolic profiles, influencing processes like cancer progression, immune response, and stem cell differentiation. However, conventional mass spectrometry provides only averaged metabolic data, limiting analysis of cellular heterogeneity. Single-cell mass spectrometry imaging (single-cell MSI) enables direct visualization of metabolite distribution at the single-cell level, revealing unique metabolic characteristics. This study performed a high-resolution metabolite imaging technique using single-cell MSI, integrating MALDI-MSI and DESI-MSI. Reporter mice expressing fluorescent proteins help analyze microglial metabolism. Single-cell MSI offers unlabeled, comprehensive detection, advancing metabolic analysis and biomedical research.
