Comparative Metabolome Analysis of Saccharomyces cerevisiae under different Aeration Conditions

The metabolic states of Saccharomyces cerevisiae laboratory strains (BY4947 and CEN.PK113-7D) and the Japanese sake brewing strain (Kyokai7) were analyzed under different aeration conditions. Under anaerobic conditions, all strains exhibited reduced growth rates, and delayed glucose consumption rates. Ethanol yield was significantly increased under the anaerobic condition, with the Kyokai7 strain showing the highest yield. Metabolome analysis revealed a decrease in the TCA cycle metabolites, suggesting reduced respiratory activity under anaerobic conditions. Furthermore, the concentrations of pyrimidine nucleotides, TTP and CTP, decreased during the anaerobic stationary phase, suggesting the involvement of oxygen in pyrimidine base synthesis. In contrast, the levels of metabolites in upstream glycolysis, the pentose phosphate pathway and aromatic amino acids increased during the anaerobic exponential growth phase, implying a shift in carbon flux towards biosynthetic pathways.
