Instrumentation Strategies for Imaging and Single-Cell Structural Lipidomics

Structural lipidomics has been developed for providing high-definition information about lipidomes with annotating lipid species at high specificity with detailed structural features resolved. We have developed the method combining photochemical derivatization and tandem mass spectrometry (MS/MS) to identify the C=C locations, sn-positions and other types of detailed structural features in lipids. While this strategy has been well implemented for large-scale lipidomics using LC-MS/MS, applications for tissue imaging and single-cell faced challenges related to the limitations due to the traditional MS/MS analysis method not capable of analyzing samples of small volumes. To address this issue, we have developed different instrumentation strategies for performing MS/MS analysis at significantly improved efficiency of using ions generated for one imaging pixel or from a single sampling of a cell. Tens to more than 100 lipid species could be MS/MS analysis for imaging or single-cell analysis. On-tissue and on-cell photochemical derivatizations were also developed to allow identification of C=C locations. MS3 of sodiated lipid ions allowed resolution of sn-positions.
