Characterization of Toxin-like Peptides in the Badumna insignis Spider Venom

Most spiders rely on venom to catch prey and to defend against predator. The venom contain salts, organic compounds, peptides and proteins. Among them, peptides are the main active components in the venom, which are expected to be applied to pharmaceuticals and agrochemicals due to their high action selectivity. In our previous study, we conducted transcriptome analysis of the venom gland of black house spiders (Badumna insignis) to search for novel insecticidal peptides. This resulted in the identification of the eight toxin-like peptides. However, there were many peptides that could not be identified based on this approach. Therefore, in this study, we determined their structure by MS/MS de novo sequencing analysis to find other toxin-like peptides in the venom. By integrating sequences obtained by transcriptome and MS/MS analyses, we were able to identify the sequences of three peptides. All these peptides share high sequence similarity to CSTX-1, an insecticidal peptide identified from Cupiennius salei. CSTX-1 was reported to show high inhibitory activity against Ca2+ channels in cockroach neurons. Therefore, it is possible that these peptides identified in this study show insecticidal activity by acting on ion channels.
