Structure Analysis of Alkaloids in Pt-coated Porous Plate MALDI Mass Spectrometry using a Spiral TOF Instrument.

When measuring compounds containing nitrogen atoms such as alkaloids using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) previously, we discovered that hydrogen loss occurs depending on the molecular structure of the compounds, resulting in observation of [M-H]+ ion.) In this study, we analyzed alkaloids using a Pt-coated porous plate, which facilitates ionization without matrix. For alkaloids such as reserpine and yohimbine, [M-H]+ ions were observed, indicating hydrogen loss. In contrast, hydrogen loss did not occur for alkaloids like harmaline and harmine. Moreover, [M-3H]+ ion of vincamine was observed in Pt-coated porous plate MALDI-MS and it was not detected in the no matrix MALDI-MS, suggesting that Pt-coated porous plate showed a kind of matrix function in strong hydrogen absorption. Addition to that, we will present the comparison of hydrogen loss reactions of alkaloids in multimatrix MALDI-MS of two different instruments of JMS-S3000 SpiralTOF MS (JEOL Ltd. Tokyo, Japan) and RapifleX MALDI TOF/TOF MS (Bruker Corporation, MA, USA).
