Establishment of 13C-metabolic flux analysis for THP-1 macrophage-like cells

13C-metabolic flux analysis using mass spectrometry was developed to investigate a metabolic alteration in immune cell differentiations. 13C-tracing analysis of THP-1, a human leukemia cell line, revealed uptakes of extracellular glutamate and aspartate as well as degradations of intracellular macromolecules. By incorporating these reactions into a metabolic model, flux distributions were calculated. The results showed that differentiation into macrophages dramatically increase the glycolytic flux, while the TCA cycle flux were moderately increased.
