Glyco-RIDGE-Based Site-Specific Glycoform Analysis of Glycoproteins: Development of a Novel Web Tool Named “GRable" and Application to Large-Scale Analysis of Mouse Tissues

For effective site-specific glycoform analysis of glycoproteins, we have developed a Glyco-RIDGE method. Here, we describe a novel web tool named GRable for semi-automated Glyco-RIDGE analysis. This tool allows site-specific glycoform analysis of sialic acid-containing intact glycopeptides in a high-throughput manner. We also present a large-scale Glyco-RIDGE analysis of six mouse tissues, in which over 10,000 site-specific N-glycoforms were identified from 800 glycoproteins. Thus, this methodology can be applied to both in-depth analyses of target glycoproteins and large-scale analyses of crude glycoprotein samples, and GRable-assisted glycoproteomics allows for investigation of disease-related glycosylation alterations and thus can facilitate the accelerated development of glycomedicine.
