フルオラス誘導体化LC-MS/MS法を用いた生体試料中シアル酸関連化合物の選択的分析
A fluorous derivatization method combined with liquid chromatography-tandem mass spectrometry (LC/MS/MS) was developed that allows the sensitive and selective analysis of sialic acids and sialyl oligosaccharides. The carboxylic acids in sialyl acids were derivatized with the fluorous amine reagent, 4,4,5,5,6,6,7,7,8,8,9,9,10,10,11,11,11-heptadecafluoroundecylamine, via amidation in the presence of 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride. The derivatives were specifically retained on an LC column with a perfluoroalkyl-modified stationary phase by means of “fluorous interaction” (i.e., the affinity between perfluoroalkyl compounds). These fluorous-derivatized analytes were detected with high sensitivity by MS/MS without matrix-induced suppression effects. This method with simple treatment enabled the quantification of sialic acids and sialyl oligosaccharides in biological samples (human urine and human milk).
