同一化合物由来の異なる衝突誘起解離プロダクトイオンスペクトルを収納したスペクトルライブラリーの有用性
Library search is a useful technique in mass spectrometry to identify compounds. The collision-induced dissociation (CID) product ion spectrum is not as reproducible as the electron ionization spectrum, and it is often the case that the same pattern of CID spectrum cannot be obtained from the same compound. We have proposed a strategy of using both database search and library search to identify various autolyzed fragment peptides derived from chemically modified trypsin in mass spectrometric protein identification. In the process, we experienced analytical examples that could not be identified even though the CID spectra of the compounds (peptides) were recorded in the spectrum library. To increase the identification rate by library search using CID spectral libraries, we performed a library search using multiple spectral libraries created from different analytical data of the same sample, and succeeded in increasing the identification rate. It is necessary to utilize a library containing a plurality of CID spectra obtained under various conditions for one compound to reduce the number of unidentified queries.
