探針エレクトロスプレーイオン化質量分析における検出感度向上へのアプローチ
In this study, to improve the detection sensitivities of hypnotics using probe electrospray ionization tandem mass spectrometer (PESI-MS/MS), the effects of addition of pH adjusters (acetic acid, ammonium acetate, carbonate buffer, and aq. ammonia) to aqueous sample solutions have been investigated. As a result, the ion intensities of hypnotics increased about 2- to 3-fold by the addition of acetic acid or ammonium acetate. Especially, the latter significantly improved the detection sensitivities compared with that of acetic acid. On the other hand, for alkali reagents, the addition of aq. ammonia led to about 5- to 10-fold improvement in the detection sensitivity, though no significant effect of addition of carbonate buffer on the sensitivity was observed. These results suggested that the ammonium ion contributed to the enhancement of ionization of the hypnotics. The optimized PESI procedure presented here would enable rapid and highly sensitive screening of adulterated samples in the forensic field.
