Development of Monoclonal Antibody Reference Material (AIST-MAB)

Extensive characterization is required for development and evaluation of antibody drug. We developed a reference material which made possible to validate protein concentration and evaluate various physicochemical properties of monoclonal antibody. The material is a humanized IgG1κ in phosphate buffer solution, whose protein concentration was precisely determined by amino acid analyses using isotope-dilution mass spectrometry. The analytical results regarding the primary structure and other physicochemical properties were summarized as technical note (case study). Both material and the analytical results are available from NMIJ.
