Glycosylation analysis of glycoengineered biopharmaceuticals produced in silkworms

Using LC/MS/MS, we analyzed the glycan structure of rituximab and etanercept produced in transgenic silkworms expressing galactosyltransferase, sialyltransferase and other related enzymes. We found that sialylated and galactosyled hybrid type N-glycans are attached to the transgenic silkworm-derived glycoproteins. Our result suggests that activity of enzymes required for complex type glycan biosynthesis may be lower in silkworms compared to CHO cells.
