Derivatization of peptides with concentrated charged droplets

It is important to derivatize peptides for improving ionization efficiency and for facilicate fragmentation. TMPP-Ac-OSu is a well-known derivatizing reagent that can modify the N-terminus of peptides, but it is easily hydrolyzed under basic conditions. Therefore, there is a need for a method to inhibit the hydrolysis reaction of TMPP and to accelerate derivatization reactions. In this study, we examined the effect of concentrated charged droplets generated by t-SPESI on the derivatization reaction and compared it to normal conditions. Results show that he hydrolysis was suppressed by using concentrated charged droplets, and the efficiency of the reaction was also significantly improved.
