Identification of Unique Metabolites in Tumor Cells for Single Cell Mass Spectrometry

Phospholipids play critical roles in transmembrane signaling processes and cellular membrane structuring. We developed a rapid method, “Live Single-cell Mass Spectrometry” for endogenous lipids in live single cells. The phospholipid profiles in the different human live single cells studied were distinctive and depended on the kind of the cells. In this study, to search for specific phospholipids specifically present in cancer cells, lipid extracts from human cancer cells and normal cells were analyzed by an LC/MS/MS. We are currently optimizing analytical conditions for detecting cancer cell-specific phospholipids at the single-cell level.
