13C-Metabolic Flux Analysis of THP1 Before and After Differentiation by Using Mass Spectrometry

13C-metablic flux analysis (MFA) using mass spectrometry was used to investigate metabolic flux distributions in differentiated and non-differentiated THP1 monocyte leukemia cells. A time course analysis of cell numbers, medium components and 13C-labeling kinetics of the intracellular metabolites confirmed that 13C-MFA could be applied for the immune cells. Flux distributions in the central carbon metabolism showed that the NADPH regeneration rate was increased by 7-fold in differentiated THP1 cells compared with non-differentiated ones, suggesting that metabolism of differentiated THP1 was rewired for immune responses.
