Development of an automatic line switcher of the desalination tube for a LC/MS

Because the detection is possible at the low wavelength, phosphate buffer is frequently used for a mobile phase for HPLC. However, it is difficult to use it for the online LC/MS analysis because the phosphate buffer is nonvolatile. We have developed a desalting tube that uses an ion exchange resin to convert phosphate to volatile ions for on-line LC/MS measurements even under phosphate buffer conditions. The time the tube can be used to remove phosphate from the mobile phase depends on the phosphate concentration and the mobile phase flow rate. For the type with an inner diameter of 1 mm, the usable time is about 10 minutes when a 10 mM phosphate buffer is pumped at a flow rate of 0.3 mL/min. If the mobile phase is pumped through the desalting tube for the entire analysis time, the tube needs to be replaced approximately every other analysis, and it cannot be used for continuous analysis. We have now developed a system that automatically switches the desalting tube for continuous analysis.
