Development and physicochemical properties of monoclonal antibody reference material, AIST-MAB

We have developed a monoclonal antibody reference material, NMIJ 6208a, AIST-MAB to address extensive needs for antibody characterization. The material is a humanized IgG1κ in phosphate buffer solution, whose protein concentration was precisely determined by amino acid analyses using isotope-dilution mass spectrometry. We also summarized the results of measurements of various properties including peptide mapping. This material can be used as a reference for antibody concentration, while provides a tool that enables a wide range of analytical techniques to be extensively compared and evaluated, going beyond quality control with in-house reference material. This material and the measured properties as the case study are currently available from NMIJ/AIST.
