Biophysical characterization of virus vectors for gene therapy by mass spectrometry
In the course of biophysical characterizations of therapeutic antibodies, mass spectrometry-based approach has been developed and now becomes one of routine techniquies for primary structure analysis. In this talk, primary structure analysis of virus vector for gene therapy will be introduced. We found that although conventional approach like SDS-PAGE analysis has been widely used for the virus vector characterizations, capillary electrophoresis together with mass spectrometry-based analysis realize accurate and quantitative characterizations for virus proteins of adeno-associated viruses, which ensure to produce the vectors with higher quality and safety. The importance and difficulty in the biophysical characterizations of virus vecter will be discussed.
