ヒト人工多能性幹細胞を用いた超網羅的タンパク質分析法の開発と糖転移酵素変動解析への応用
Protein glycosylation is one of the major post-translational modifications. Quantitative studies of glycosyltransferases have a great impact on understanding biological function of glycosylation. The data-independent LC/MS/MS (DIA-MS) method is a powerful tool for quantification of proteins; however, the DIA-MS requires a comprehensive library, including low-abundance glycosyltransferases. In this study, we performed 1) a sample preparation to create an ultra-comprehensive protein library using hiPSCs, and 2) quantitative analysis of glycosyltransferases during differentiation of hiPSCs into neural stem cells by DIA-MS. We found the changes in the levels of several glycosyltransferases associated with O-glycosylation.
