探針エレクトロスプレーイオン化タンデム質量分析(PESI/MS/MS)による園芸作物におけるアントシアニンのハイスループット分析
Secondary metabolites are important for traits of horticultural crops; color, aroma, taste and functionality. High-throughput analysis of these metabolites have been required at breeding field, distribution and consumer market. We applied probe electrospray ionization (PESI) to evaluate the quality of plants, especially horticultural crops. PESI does not require pre-treatment and separation of samples, and so it is suitable for high-throughput analysis. In this study, we targeted anthocyanin, one of the most major pigments in fruit and vegetable. Nearly 70 kinds of anthocyanins were detected in approximately 3 min at selected reaction monitoring mode. Tandem mass spectrometer (MS/MS) could distinguish the fragments of anthocyanin and flavonol, adequately. Probe sampling, the simpler method sticking a probe directly to sample, could detect anthocyanins specifically in micro area scale. In strawberry, fruit including seed (achene) and pulp (receptacle) accumulated different molecular species of anthocyanins. PESI/MS/MS could be utilized to characterize the profile of anthocyanins, and to compare the contents among cultivars quantitatively.
