in the present study, head lettuce (Lactuca sativa L.) grown in an artificial climatic chamber was investigated and then the primary metabolites in leaves were measured using CE-MS to analyze the growth conditions under which crop functionality can be enhanced in a closed environment. Lettuce plants with 1,000 ppm CO2 and/or nutrients (Hoagland's solution) supply for two weeks were analyzed. The results showed that the growth of lettuce grown in the high CO2 condition alone was inhibited, while simultaneous treatment with high CO2 and nutrients increased fresh weight by about five times that of control plants. The measurement of primary metabolites revealed that many amino acids such as glutamate and aspartate were significantly increased in the lettuce leaves treated with both high CO2 and Hoagland's solution. Moreover, a significant increase was found in fresh weight of lettuce red LED irradiation for a week. Asparagine, arginine, and branched-chain amino acids tended to be increased in the red LED-exposed plants. Simultaneous treatments of monochromatic LED, high CO2 and nutrient formulation would improve the accumulation of amino acids, and likely reduce the inorganic nitrogen sources in crops.
