UHPLC-FTMSによるヒト前房水を用いた緑内障バイオマーカー探索研究
Glaucoma is the primary cause of blindness characterized by progressive loss of retinal ganglion cells and irreversible visual deficiency. The candidate of lipid biomarkers previously identified with aquas humor derived from the glaucoma patients1). Although we also demonstrated the phospholipids increasing early in the retina of glaucoma model mice2), the detailed components of the lipid species were still unknown. We therefore characterized the lipid species in human aquas humor and searched prognostic biomarker the by means of the lipidomic profiling. The lipid species were extracted from 83 samples, including primary open-angle glaucoma (POAG), pseudoexfoliation syndrome (PE) and normal tension glaucoma (NTG) by methanol precipitation and Bligh & Dyer extraction methods, and analyzed by UHPLC-FTMS. The variation of lipid species in POAG, NTG, and PE could be observed by the multivariate analysis, such as principal component analysis and heretical cluster analysis with volcano plot analysis due to glaucoma increase in an intraocular pressure-dependent manner, suggesting that the detailed the lipid profiling may give us the novel biomarker of the glaucoma patients.
