GC-MSを用いた好中球様細胞HL-60の13C代謝フラックス解析法の構築
13C-metablic flux analysis using mass spectrometry was employed to investigate metabolic flux distributions in undifferentiated, differentiated, and LPS-activated HL-60 myeloid leukemia cells. First, the metabolic model was determined by 13C-tracing. Glucose and glutamine, as well as aspartate, and glutamate were incorporated into cells. The metabolic flux distribution calculated using the constructed metabolic model showed that the level of TCA cycle fluxes was constant, while differentiated HL-60 decreased the level of glycolysis flux compared to that of undifferentiated HL-60. LPS-activated differentiated HL-60 cells showed that increased levels of flux of glucose uptake, pentose phosphate pathway and NADPH regeneration.
