Co-Fractionation MSによる細胞内巨大タンパク質複合体の解析
Several different methods have been developed to analyze protein complexes. In addition to Affinity-Purification (AP)-MS and proximity labelling approaches, e.g. BioID and APEX, Co-Fractionation Mass Spectrometry (CF-MS) analysis has been recently introduced for the systematic characterization of cellular protein complexes. Using uHPLC-based Size Exclusion Chromatography (SEC), we have shown that even very large cellular complexes, including ribosomes and polysomes, can be separated efficiently by uHPLC using SEC columns with a pore size of 2,000Å. Here, we apply this technique to isolate and characterize intact nucleolar pre-ribosomal particles from human cells.
