ヒト組織のハイブリッド・イメージング:疾患脳の分子・原子アトラスへ向けて
In our body, various molecules constitute tissues and organs, and work efficiently to fulfill their physiological function in a coordinated and dynamic manner. When the molecular coordination breaks down for some reason, its pathological modulation appears as disease. Therefore, it is important to obtain as much information as possible on the quality and localization of all molecules and atoms on the body tissue and to analyze them precisely in an integrated manner. In this talk, we will present about the integrated analyses of the same tissue from a human brain by applying various analytical methods, combining the conventional histological analyses with MALDI (matrix-assisted laser desorption/ionization)-IMS (imaging mass spectrometry), SALDI (surface-assisted laser desorption/ionization)-IMS, LA (laser ablation)-ICP-MS, and PIXE (particle induced X-ray emission). We will discuss the current situation and issues, and introduce our efforts on integrated omics analyses against human disease brains in order to elucidate the disease mechanisms. Our studies show mass spectrometry is a promising approach that can deepen our knowledge about the localization of biological molecules in human brain tissue during pathogenesis.
